I tried (in terraria) to remove the sound files (the .xnb files only), leaving the sound folder empty. It turned out, that it CAN. The game ran completely fine without them (but of course with a sound).

So my question is, what can block a game from reading a .xnb file ?
It might be some corrupted files in your Windows installation. I don't see any other fix than doing a clean install (reset) of your pc

Try to follow this guide, it might fix your problem, or run the windows troubleshooter.
